No, any colors you currently have in your Kubrow color palette can be used in any of the three slots. To expand your color set, you have to go to the Market > Equipment > Kubrows. Most of the colors are available through platinum only though, but the Basic Gene-Masking Kit is available for 100k credits and that has 5 colors in it. Hope this answers your question.
