I could see two possible reasons for why they are giving us only twelve notes instead of sixteen.  The first is the UI getting too crowded with so they capped it off at twelve notes.  The second being that they may not want to overwhelm users with too many options.

Maybe what they could do, to avoid overwhelming users, is start with a set amount of notes and as we level up the powers/warframe we unlock additional notes until we reach sixteen notes.  For example, we start with four notes, and with each rank of our fourth, or third, power we unlock an additional three notes.

See how the first column is 16 A's stacked on top of each other? Each A in that column is a beat.

How it works is that yellow line radiates from the middle, and as it reaches each note, that note is played. So the lowest ones will play first, and as the yellow line radiates outwards, each note it touches will be played until it reaches the highest note in the column, which will be the last note.

Sorry if that makes zero sense.

The columns of letters each represent a note at different times in the song that plays on a loop, which begins from the lowest part of the columns and goes up.

The columns are divided in 16 beats. The position you turn the note on determines when it plays in the loop. If you just turn all the beats in a single column you'll just get a single long note lasting 16 beats.

Watching the demonstration at Pax Aus really helps understanding it, if you haven't yet.

Ah, I got it, I was confusing the lines with the columns.

But seems to be a shame if we are unable to make a full music, or a choirs at least, I would have liked to make a "Cara Mia Addio"

Yes, you can have multiple notes sound on a given beat. With less than a whole octave to choose from though, your options for harmonization are pretty limited.

Also, at Pax AUS, they mentioned they might limit the maximum notes you can have in a loop to prevent troll-ish players from creating a cacophonous mess by having every note play on every beat.

From the demo Steve gave at PAXAus, you will be able to have sustained notes up to fifteen beats long or continuous.
